I am not a fan of the NAD boosters. There may be people they help, but I don’t see them as a tool to improve mitochondrial quality even if they might temporarily improve some function for some people.
I just came across this thread and wanted to share my improvements with SS31. I do have proven mitochondrial DNA damage on biopsy so my case may not be applicable to others.
I notice improvements that scale with dosage and am currently on 16mg/day with a goal to titrate to the full 40mg/day. Others have speculated that the higher dose is only needed in Barth Syndrome but that’s not how the dose was calculated. In phase I trials, patients were given it via IV and plasma levels were measured. Those that received 0.25mg/kg for 2 hours for a total of 0.5mg/kg reached steady state plasma levels. That was converted to subcutaneous dosing via its known pharmacokinetics and that is how they came across the 40mg dose.
For me I have a reduction in heart rate, improvement in HRV and increased deep sleep time since being on SS31. I have had no increase in my pitiful low step count but that may change as I titrate up to the maximum dosage of 40mg/day.
As for sourcing why does everyone buy these kits from resellers? I buy directly from Chinese lab around $250/gram and typically in a group buy with others so we test it. Seems the purity is always high (99%) but the peptide content is always between 70 - 80% and the rest is the salt. So in order to get to the 40mg of active drug per day one will have to inject 50mg from Chinese sellers. The labs are not a secret as I have seen one of them mentioned in this thread already.
Anyways is there anyone else here who has seen such great improvements from this peptide? Also does anyone know of any other peptides or drugs I should try to help reduce my high mtDNA deletion?
Welcome to the forums, and thanks for sharing. Can you tell us how long you’ve been taking SS-31? What doses have you used and for how long (what has been your dosing protocol during the entire time you’ve used it?). Are you working with a doctor on this, or just biohacking?
I have a mitochondrial geneticist I see but it is not prescribed by them. I was open with them and telling them I am sourcing this myself and they seemed okay with that since they have no treatment options besides vitamins to prescribe.
I previously took this when I first got diagnosed a few years ago but didn’t stick it. Since resuming I have been on it for about 3 weeks now. I started at 1mg and have been increasing by .8 - 1.6mg per day. Over the three week period my resting heart rate is lower by 3 bpm, my HRV is up by 4ms and my deep sleep is up by 20%. My step count is still under 1000 steps a day due to my disease severity. These are not dramatic results I understand but I am only on for a short period of time and have not yet reached the targeted dose.
I tend to be sensitive to medications given severe dysautonomia which is why I am doing a slow titration.
I’d check out the thread on ergothioneine if I were in your situation, with the caveat that you may need to look at some genes and/or do some post-supplementation testing for TMAO to help ensure it’s safe to take long-term.
I felt like SS-31 helped with my ADD symptoms….the focus and motivation to get things done.
Having said that, I can’t be sure it wasn’t placebo or something else.
I stopped taking it when ‘life happened’ and decided to take that opportunity to take a longer break with the goal of starting again to see if I notice a change. I’ve had a couple of false starts, but I have been very lazy and have not had the oomph to inject daily … ha, if only I was taking SS-31 :).
Now I’ll know to check out my oura stats when I start up regularly again to see if I notice anything.

I have an Apple Watch and have just looked at the monthly view of how they have changed. It’s very small changes and could of course be secondary to something else.
I definitely understand the stopping and starting. It’s so hard to commit to something long term especially if it’s unclear if it’s helping or not and the barrier is high (sourcing, injecting etc).
I didn’t include subjective changes in the initial comment because I wasn’t sure how to really evaluate them. I will say that I feel like an improved mood on it with less lows and less anxiety. But that could also be because I’m sleeping better and have less pain.

Is ergothioneine use related to TMAO? Caught my attention because I take quite a bit since I get it just for some effort kind of like gardening. And my TMAO was very recently tested off the chart High. I was doing several dumb things and it’s probably just my gut microbiome. I’ll keep testing. Just wondered whether you know something my AI doesn’t.

But, the other thing to be potentially concerned about is the genetic variant (which appear all too common) that increases ergothioneine uptake into tissues and could make it potentially harmful if supplemented in large amounts.
I injected 20 mg x 10 days + 10 mg x 15 days of SS-31. I did not notice anything. An optimistic hypothesis is that my mitochondria are in too good condition to make any effect noticeable. I missed making any tests.
I might consider another try. 40 mg per day as per the dose for Barth and simultaneously taking intranasal Orexin-A on the possibility that a lack of this could prevent effects of SS-31, as detailed in the thread on Orexin.
I would think at the doses you have tried that your conclusion is correct. Your mitochondria are likely healthy and probably won’t benefit from going even higher.
To be honest, I find that hard to believe. Is it even possible for anyone over the age of 65 or 70 (Ulf, I forget how old you are exactly) to not have significantly degraded mitochondria? Ulf, no doubt about, you are in great shape and doing well (much better than most people your age, or even much younger).
But… I would still think that even for people in good shape the mitochondria would be significantly degraded, and if their cardiolipin membranes were returned to functional youth, that you’d feel a significant benefit.
I wonder if the dose might be too low? Or perhaps there is a “wrong order” in the peptide delivered?
