Rapamycin Dosage - weekly vs. daily

I’ve always wondered about the recommendation to take the weekly rapamycin dosage all in one day. What is the rational for this? Why not take it throughout the week (e.g., daily) to maintain on constant dosage rather than a big spike that diminishes throughout the week? I seem to recall that Matt Keberlein did an experiment which showed he was nearly at the zero level in his blood after 7 days. Sorry if this has been discussed before.

@John_Hemming has a theory that the trough is as important as the peak.

Other than that, I don’t think we have much data on daily vs weekly.

I am also running on about every 6 weeks or slightly more. I wait for 6 weeks and then decide on a good time to go for it.

I believe the general thinking on this is you want to try to mimic the mTORC1 cycles of youth (sometimes on, sometimes off) not just block mTORC1 all the time. This is from Brian Kennedy in his discussions on rapamycin.

Also - you want to avoid mTORC2 inhibition - so taking breaks helps guarantee that.
