Out of curiosity, why do you take GFJ after taking Rapamycin? Shouldn’t you take it all beforehand as you are trying to knock out the enzymes before your Rapa is absorbed?

Dose thirty 8mg{2mg X 4, Dr. Reddy’s Brand] - 09/11/2022

Had ¾ of a large organic GF, waited approximately 45 minutes

Took with wild sardines in OO.

Yes, I still took my weekly dose.

Last week several time start adding /using my rapamycin/DMSO{amount is around 1.5mg per ml of rapamycin in DMSO, diluted with 0.9 sterile saline, DMSO around 30%] topical on skin dip finger in solution rub on inside wrist radial artery area. Also around my left front knee area, under the knee cap. Also a dab behind each ear and rub some on my forehead hair line area.

I do not think it was more the 1ml total liquid amount used.

Late Friday have a very small ulcer in my gum upper inside lip area side where the gum and inner lip join. First time since talking rapamycin, started on 02/20/2022

FWIW, the DMSO is transporting the rapamycin very well through the skin.

This was going to be the solution for intranasal at a lower % of DMSO, I had an issue while mixing, when I added the 0.9% saline saline had instant crystallization develop in the solution. So I increased DMSO amount, just to use topical on skin area.

Still working on a intranasal solution mix.

Instant crystallisation when diluting a DMSO/Rapa-mixture with saline (in his case I mean to remember it was demineralised water), is also what MAC mentioned. This is a pity, as there doesn’t seem to be an easy way to avoid using 99.9% DMSO.

Transcutol is great for the topical rapamycin skin cream:

Where you can order / buy Transcutol :
LotionCrafters: Transcutol / Ethoxydiglycol
Laballey.com: Transcutol / Diethylene Glycol Monoethyl Ether

I am envious that you can go up to 8 mg + GFJ + Fat source and experience no side effects! Humans truly are different creatures physiologically!

I hope that we can all get the same benefits though…

I can confirm that Dr. Green still recommends 6 MG, still prefers weekly dosing, and does not give any recommendations about taking with food. I know he would disagree about adding GFJ (ditto Matt K) but that never came up. I have started taking mine with fatty foods based on what I have read on this site. There may be benefits to a higher dose, but an improved immune response does not seem to be one of them. Five MG performed much better than 20 MG.

Zazim, could you please elaborate on this statement? How is it much better?

It’s all available in the Mannick paper though they used Everolimus not Rapamune which is ~80% as effective:

And like all rapalogs ridiculously more expensive, but the results are valid for our conversation. Peter Attia has a good podcast with the lead author on this paper and also one with Joan Mannick.

Actually everolimus is now generic and about the same price as rapamycin. Easily available from the regular Indian online pharmacies.

Yes, but resTORbio seems to be focusing on RTB101 which isn’t generic.

If the Mannick paper (which showed mTOR inhibition improves immune function in the elderly) is being used as the framework for rapamycin dosing, shouldn’t we be using bi-weekly dosing of sirolimus instead of weekly, given that the half-life of sirolimus is roughly double that of everolimus (aka RAD001) used in the Mannick study?

I tried Dr. Green’s formula for rapa cream. (.045%) Mixed two batches of Cetaphil cream 200 grams + 90 mg. Rapa, and I liked the results. This is what Green used on himself with good results on sun damage to head and face. This time around I am adding propylene glycol to enhance absorbtion. Transcutol is controlled where I live in EU. Propylene glycol is used extensively in cosmetic industry, to promote absorbtion, and has a safe profile. So my new formula is 200 grams Cetaphil cream + 90 mg Rapa powder disolved into 2 oz. propylene glycol all mixed well. After mixing, I could have probably used 3 oz Propylene glycol instead of 2. (experimeting on how much Propylene glycol to use per 100 grams of cream) I am expecting very good results. I should say that using tablets can be expensive for some, but powder is the best way for all rapa uses, oral, IM, IN, and topical.
