What dose did you use at the highest point?

What is new study showing best to take metformin in morning?

8mg once a week… built up slowly over 2 months and I persisted with it

Are you still taking rapamycin, or are you taking a break now? Will you take it again?

@Sfcomms & @nym Very interesting regarding the improved oral health. What dose regime are you currently on? Curious also what brand and manufacture of rapamycin you use if that can have any impact.

i’m on 5mg weekly now but started at 1mg for a couple of weeks, then 2mg for a couple of weeks, then 3, you get the idea. i take sirolimus pills. i noticed dental improvement by the time i got to 4mg so around 2 months. i don’t have periodontal disease but deep pockets and my gums bleed when i floss, which i do daily. it’s genetic. i noticed it and then my dentist confirmed. we are giving it a couple more months and will take for,al measurements and x-rays to compare.

my 1h levels were 4.5 and 2.2 at 48h (with 4mg). i would invest in testing twice to see where you are and make sure you are washing out (or have anything to start with). keep in mind that there seems to be gender difference in bioavailability levels. i’m female
cheers

I have noticed the last two times that I’ve taken Sirolimus (every other Saturday) the first week following I feel run down and achy, around 8 days later I feel very good and strong.

Last dose taken; 5mg with grapefruit juice 1 hr before.

Anyone else?

I think you should include something like brain fog. I just couldn’t remember my nouns. People’s names or the names of things I want to talk about. Really embarrassing and inconvenient and Rapa knocked it right out.

-I’d like to know the diet of users here, like low carb, keto, SAD, low fat, low glycemic / slow carb (me).
-age ranges
-taking Rapa on weight training days vs rest days.

I’m also wondering about any correlation of dosing levels to side effects and severity of side effects. Maybe in the next survey I split the respondents up into dosing tiers - e.g. 1 to 5mg/week, 5 to 10mg/week, 10 to 20mg/week, 20mg to 30mg… etc. - then ask about side effects for each group. We should see a dose/response relationship in the side effects.

Sounds like a good question - you seem to know something about these diets, can you draft a question I can use, and provide rough guidelines so people can self-identify with a specific group. If we just say “low carb”, people’s interpretations on what “low carb” means may really muddy the data.

6mg once per week, using Sirokem from Kalchela Medex - generic from India. Not sure of the quality, but it’s having some effect, judging from the improved oral health alone.

I would like to see the frequency and dosage results corelated. I’ve been doing 6mg/wk for a year. Just switched to 8mg/10d. Looking for any changes.

Somewhat like… me I did 6MG for one and a 1/2 years… then started 8MG to 10MG for 2 months and then started the 6MG with grapefruit juice to date… yesterday. Gives me 30ng /mL in my blood.

Me too! I feel great after a week off but I am fatigued for about 5 days.

There is a connection between rapamycin, “brain fog”, Covid, aging, cardiac fibrosis and something called the NLRP3 inflammasome.
Severe Covid or aging can overactivate NLRP3. Conversely, inhibition or knockout of NLRP3 can result in reduced cardiac fibrosis, an increase in autophagy and NAD+ levels.

So, if we look at things that block or inhibit NLRP3 we might find things that reduce the severity of Covid, counter aging and brain fog (closely allied to Chronic Fatigue Syndrome) and reduce cardiac fibrosis.

The list of NLRP3 inhibitors includes (but is not limited to):
Glucosamine
Metformin
Salvianolate
Triptolide
Beta-Hydroxybutyrate
Rapamycin
Hydrogen

This is an amazing explanation. I’ve been trying to read about it all morning. My dad had this problem as well. His brain was working just fine but he couldn’t find the words he wanted. We must have gotten the overactive NLRP3 gene.

I’m taking glucosamine daily and Rapa every 2 weeks. I’ve been putting off asking my doc about a low dose of Metformin. My sugars are so normal that he will drag his feet I think.

Keto works great for me but I eat enough of veg. and berries and they have so many carbs that I don’t spend any time in true optimal ketosis. Even when I was very strict I really couldn’t do it. I did just order Juvenescence to see how I like it. I used to use a lot of exogenous keto salts and thought they worked, but I don’t know how well. They helped me stay alert when keto on hard work days. So maybe the beta-hydroxybuterate from the Juvenescence will keep my brain working this fall.

This could help with heart disease too!

Thanks, Doc

I like this idea - but I’m unsure on how to implement this in a survey like this (given the software limitations… very simple polls (no branching, conditional questions, etc.). Any suggestions greatly appreciated.
