Contraindications with Rapamycin and Modafinil

Has anyone taken Rapa and moodafinil on the same day and if so, have you had any contraindications. I take one modafinil twice a week for recreational use and I love the feeling. In fact I have introduced it to my husband and it motivated him to do all kinds of garage tidying and backyard planting . I have not taken Rapa on the same day in case it reduces the impact. Any thoughts on this?

As a matter of fact I did this just today. 200 mg of modanifil early in the morning before drinking grapefruit juice an taking 8 mg of rapamycin two hours later. I have been doing this for some time without noticing any adverse effects. Though, I would say if you have just started using modanfil or rapamycin maybe I wouldn’t take them on the same day. I was very sensitive to modafinal when I first started taking it.

I envy those of you who can take such doses of modafinil (in the AM) and still sleep that night. I think my max is about 25mg. sigh.

I completely understand, I take 200 mg only twice a week because of the sleep problems, in fact , at 1 am this morning I got up to take a melatonin because I couldn’t sleep, however, the time before, I took an antihistamine before bed and slept 8 hours so you might want to try that . I have a great day on Modafinil , I’m so happy I found this on here

Thank you Charles, it’s because I read one of your messages on here that I learned about Modafinil and I cannot tell you how grateful I am. It’s a gamechanger. I only take one twice a week so that I can sleep ok. I will now be taking it on Rapa days if needed after your response , thank you

Thank you Lessa, one of our members takes modafini on the same day as his Rapa so it sounds ok. Do you have to take a sleep aid on Modifini days?

I can’t take melatonin at any effective dose. 1) I don’t clear it for 24+ hours, and 2) and at higher doses, it makes me feel like my skin doesn’t fit, again, for 24+ hours. It’s most unpleasant. Also, 3) even at high doses, it doesn’t put me under in sleep; it makes me feel drugged and not able to get deep. Melatonin doesn’t work for everyone.

That’s a shame, have you tried Amazon allergy relief, , it works great for me and no drowsiness the next day

I take Rapamycin low dose 1mg intermittently to tame my overactive immune system which brings about eczema and psoriasis with reasonable good result.
I also take 10m Vyvanse now and then on days I need extra mental acuity.
I have not noticed any “contradiction” but I wonder if Modafinil is better than Vyvanse for people do not have ADHD?

I have taken them on the same day several times before, including yesterday actually (about 30 minutes apart). I never notice anything subjectively one way or another. I don’t think it would be a problem.

You asked about a sleep aid and unfortunately, 200mg Modafinil first thing in the morning still interferes with my sleep. It is really unfortunate because that is the reason I don’t take it more often.

The reason I am now taking a 200 mg dose in the morning is that modafinil is like many nootropics (psychostimulants). You soon become adjusted to it, and it loses its effect. I started out with 50 mg in the morning, and even that low amount made me feel wired, but it did not affect my sleep that night.

I have titrated up to 200 mg in the morning, the most often prescribed dosage, but now I barely even notice it. When it’s working, it gives me a mental energy boost all day long. I have always felt sleepy in the early afternoon after lunch, and I am prone to napping. Modafinil eliminates the need for naps. In that respect, it still seems to be working. Modafinil is probably best used intermittently on those occasions when you didn’t sleep well.

I am going to try alternating weeks between modafinil and sulbutiamine.

Good discusion here re modifinil. My wife takes 50mg before 8am. I suggest anyone taking this iit should be as early as possible. <9am.

One coach uses moda for BDNF braiin training and doses: M=25mg, W=50mg, Fr=100mg. His logic is the brain trains at each BDNF level till its new base line is higher. TBD

My use is intermittent at 100mg <<9am. AND 200mg the morning of any dangerous activity like long distance driving, construction project etc. But not daily and if frequent use I try to stop Sat/Sun.

I never considered rapa / moda interaction or reduction in rapa’s efficacy. I can’t imagine…

Thank you Curt , my tablets are 200 mg, I started in 1/4 of a tablet but didn’t notice any effect, then I tried half a tablet, very little effect , I take 200 mg twice a week and I haven’t had any reduction in benefit. I take a antihistamine on those nights to help me sleep and it works, I find that the following evening I’m ready for bed much earlier and normally get a full 8 hours sleep , I’m a very mature female :grin:

@Deeinflorida BTW many drugs are anti-chologenic (??) strips the brain of the essential nutrient choline. Antihistamines, Tylenol PM etc etc are very bad for brain health, memory, brain longevity.

I have sworn off tylenol PM and other drugs that are anti-brain health. AND take many forms of choline, PS, alpha-GPC, etc.

Please reconsiider your use of antihistamines et al.

You may not benefit from modafinil and may just need to stop and move on.

But being 72yo and focus alot of my nutricals toward brain health, brain performance I can offer this partial list of things to add to your daily list:

  • astataxin (??) many sources
  • phenylpracetam
  • aniracetam
  • noopept
  • ketones; Ketone-IQ, goBHB
  • higher protein diet. Protein feeds nerve/brain
  • Plasmologens; Take both of the forms from prodrome.com
  • PRM, pro resolving mediators; / quality fish oil. I like: Big Bold health: omega 3.
  • Another PRM variant worth taking: SPM supreme by designs for health

A keto genic diet. As we age our brains become more and more benefiting from ketones >>1.5 best over 2.0. We eat carnivor and use exegenous ketones liberally to get circulating ketones up without haveing to fast or drop carbs ultra low. BUT fasting, strength training are very useful to aging health.

Seems to put me to sleep faster and then I wake up around 2am and can’t get back to sleep. I hate it.

I took CBD oil and 2 sleep supplements last night and that helped but I did wake up a few times, I hate that too
