Took too much Rapamycin so wish me luck

It was a bit early this morning and I wasn’t fully awake so I miscalculated my dose, 16mg with grapefruit juice, so wish me luck. So far I notice nothing and by Wednesday most should have cleared my system though

Stay close to the bathroom, you will probably get diarrhea.

Good news is that it most probably will not kill you. But that is a pretty high dosage. Be careful!

good thing is the really high levels only last for around 3 days, after 3 days my levels will be lower then I had initially intended (wanted to take 12, previous highest dose 10mg with GFJ)

If my experience can serve as a guide, you probably shouldn’t worry.

I took 12 mg with grapefruit juice every 14 days for a year with no side effects - peak was 68 ng/ml and trough was 1.9. That’s a little less than your accidental dose, but it was long-term.

Highest intentional dose of rapamycin ever recorded (suicide attempt) was a little over 100mg and that person survived without issue, so I am not too worried. Still it was higher than I wanted to (I am also an ultra fast metabolizer of rapamycin)

I’ve ever taken rapamycin 30mg + grapefruit 400ml, I experienced headache at the day 2-4, diarrhea at the day 14, acne within the day 2-14. The Headache and the diarrhea are mild and tolerable, but the acne is very painful and intolerable.

Quite possibly you took the highest dose of rapamycin ever recorded then… ( don’t know whether or not the person that attempted suicide with rapamycin too GFJ)

I have taken 20 mg with EVOO and GFJ before and experienced mild diarrhea.
Please share your side effects. I also experience mild euphoria on the second and third days. The euphoria may have been coincidental as I never experienced it again.

Nothing out of the ordinary after increasing my dosage so far, just tired and weak like before although it seemed to last a while longer

Do you have any idea why that might be? I have had a little trouble sleeping on the same day with a large dose, but I have never noticed any weakness or tiredness.

I just took my largest dose 4 mg + GFJ + EVOO. I thought I didn’t have any side-effects, but then…

Last night:

  1. Had a hard time sleeping even though I took the Rapa at 1 pm.
  2. Bad diarrhea twice
  3. Euphoric fatigue (feeling of well-being yet tired)

I enjoy #3, so that’s more of a benefit. I am used to #2 from my heavy metformin days. #1 was probably the hardest, but not bad compared to some other days.

At least no canker sores, rashes or acne (yet).

Up to 6mg in today’s dose. I’m still not experiencing any affects at all from rapamycin.

Will probably increase to 7mg next Sunday, then keep it at that dose level.

How did you conclude that you’re an ultra fast metabolizer of rapamycin?

Interesting. Could you share which genome sequencing test you took and which variants were indicative of the fast metabolism? Did the test give you the raw sequencing data which you then uploaded to a website to get information on the rapamycin metabolism, or did you look for variations in CYP liver enzymes yourself?

I did dante, I got the pharmacogenetics report for free with the 30x wgs. I can’t look up which variant I have today

I recently dropped metformin because I can’t find any studies that it is beneficial for people over 80 yrs. old.
In any case, since I have stopped metformin my digestive system seems much happier.
My suggestion is: On days when you take “4 mg + GFJ + EVOO” don’t take metformin.
I have had zero bouts of diarrhea since I stopped metformin.
Also, take your rapamycin early in the morning to minimize sleep effects.

Thanks for the advice. It seems I have the same stable of side effects and taking more Rapamycin just makes them worse. But then there are treatments to prevent some of them. The most bothersome is the not sleeping well one. I may just back off down to 3 mg, but I was hoping to try 5 mg first.
