Prolonged tadalafil exposure promotes skeletal muscle myogenesis and regulates early myogenic markers activation in C2C12 cell lines

Conclusions: Tadalafil promoted a highly proliferative state and preventing premature differentiation. These findings suggest its potential use as an adjuvant pharmacological therapy in the treatment of muscle-wasting disorders. However, the potent improvement of muscle regenerative machinery raises critical concerns regarding its potential misuse as a performance-enhancing drug in sport, highlighting the need for evaluation by anti-doping agencies.

This is in cell lines in a dish (in vitro). And what is “prolonged exposure” here? It’s 7 days… OK. But what is the ultimate long term effect in the human body for overall health outcomes, especially if you don’t have any muscle wasting? At least steroids reliably promote muscle growth in vivo, and depending on dosage there are no notable negative side effects at least short to medium term - coming up with drugs that promote muscle growth is something gym bros have done for decades… but impact on overall health… Anyway, they themselves see potential for abuse in exactly this context, so maybe bodybuilders will get excited. But for biohackers, this petri dish effect is early days indeed. YMMV.

Can’t access the paper yet (I will try to get it over the next week or two), but we don’t know dose levels, and it was in cells… so not very translateable to humans, at least right now. I’m not too worried about this but will learn more and read the paper when I can.
