I have been taking melatonin at various high doses for many decades, and I also exercise regularly. I have detected no sarcopenia at age 83.
As I posted earlier, if I could only take one supplement, it would be melatonin.

Agreed… Mel is currently the MOST underrated supplement the need for which exponentially increases with age

The free “Supporting Information” document contains the answer: "The melatonin content in the rodent chow (50 mg/kg) was determined based on the average daily food intake, number, weight, and age of the mice, ensuring each mouse received 10 mg/kg of body weight daily. This dosage was chosen based on our previous studies, which demonstrated its efficacy in affecting all subcellular compartments (20-22, 28, 29). The concentration of melatonin in the pellets was previously verified by HPLC, where the theoretical concentration (50 mg/kg rodent chow) was compared to the measured concentration (47.2 ± 2.0 mg/kg rodent chow) (21). "

Using the FDA guide, 10 mg/kg/day in mice => 0.81 mg/kg in humans => About ~50mg/day for a 60-kg human.

I bought the paper but it doesn’t even contain that information… Anyway, the figures:

The really high human doses have tended to be a last ditch attempt to deal with cancer. However, because there is uncertainty I personally have decided to err on the side of excess (for which there appears to be no identified downside).

What’s the highest dose you tried and what is your current regimen? Immediate release or prolonged/extended release?

How much melatonin do you take and when?

I tend to follow whichever way the current wind is blowing.

I started melatonin in 1985 to counter the effects of jet lag.

The dose I started with was 3mg. I have never taken less than that daily.

The highest dose that I have taken recently is 2 grams, once at bedtime and once again upon waking.

I find it interesting that taking 1 gram of melatonin upon waking had no effect on daytime sleepiness.

Currently I am taking ~800 mg before bedtime. The 800 mg has no special significance other than the amount in each capsule that I fill myself with. The main reason for filling capsules myself is that I can get pure melatonin with no fillers. 800 mg is close enough to 1 gram, which I was aiming for.

The reason I take 1 gram is for its possible cancer prevention benefits, and as @john_hemming says, there seem to be no downsides. At least I have never experienced any negative consequences.
The fact that it might help with sarcopenia was news to me.

There are many studies of shift workers where the natural production of melatonin is inhibited and cancer rates increase.

I have recommended high-dose melatonin to my daughters and other female relatives for the possible prevention of breast cancer. The amount of melatonin they are taking is greater than 20 mg. My daughters have experienced a dramatic decrease in menstrual symptoms and the symptoms of menopause.

An effect of melatonin is to delay development. Menopause is development.

What is unclear is whether melatonin can reverse development.
