Intro and side effects - knee pain and tingling in foot

My main hope for taking rapamycin is that it reduces inflammation and slows down neurodegeneration. I had chronic back pain for years and guessed it was due to inflammation, so I followed a ketogenic diet hoping it would resolve that issue, and miraculously, it did, but I’m not able to stay on the diet long term because I become weak. I don’t understand how many similar inflammation pathways rapamycin activates but I’m hoping it will do the same without making me weak.

I’m also encouraged that rapamycin could slow down neurodegeneration since I have glaucoma. I’m hoping rapamycin will help resolve my allergies, occasional mild ashthma, chronic rhinitis, flushing and hives (which is probably due to high histamine levels – my dad had the same issue and a doctor told him he had high histamine levels). After my dad starting taking a mild immunosuppressant for cancer those issues resolved.

I took my first dose last week of 3mg (I’m a 57 year old 120 lb female), and the 3rd day I had intense tingling near the ball of my left foot. It spread a tiny bit to my right foot. It didn’t hurt but was very alarming. Like my foot fell asleep but a lot more intense. I paced around my house for 15 minutes hoping it would dissipate, which it did within a half hour. The 4th day I woke up with my arms feeling a very slightly numb like the circulation had been cut off, and I had throbbing pain in my left knee (and minor pain in right knee) for 70% of the day, which walking, stretching, naproxen, and sitting with legs straight wouldn’t resolve. I’ve pretty much never had knee pain or tingling so this stood out as unusual and correlated with taking rapamycin. I did have about 2.5 glasses of wine throughout the 4th day (which is unusual for me, I don’t drink often and rarely more than 1 drink). I do wonder if the alcohol had anything to do with it not going away, but I woke up with the issues. Thank goodness the knee pain resolved on the 5th day, because otherwise I’d hesitate to keep taking rapamycin, assuming that was the cause.

Also have bitter taste in mouth often but seems most likely it’s the new glaucoma drops I just started taking.

Has anyone had similar side effects or similar issues that rapamycin helped resolve? I did see a couple of posts about knee pain.

My own experience is that rapamycin resolved joint pain. At 83, I am pain-free as long as I don’t do anything to aggravate my lower back. I had a lot of knee pain in my right leg from tweaking while playing tennis. All joint pain has been resolved since I started rapamycin.
To be fair I also take a Boswellia Serrata supplement which is supposed to reduce joint pain, especially knee pain.
I may be atypical because I suffer none of the usual side effects from taking rapamycin. The only side effect of rapamycin I have experienced is diarrhea from taking too high a dose (20 mg). I recommend taking a Boswellia Serrata supplement if you suffer from joint pain. There is a fair amount of evidence that it is effective.

Thanks, I might consider trying boswellia, or filing away the info for later, especially since it might help asthmatic symptoms too.

My knees and I thank you for your recommendation. I had lost the ability to do unweighted squats. Strangely I could do 50lb squats with no pain. Started May 28 with 5000 mg Boswellia, and today I can now do 20 squats with no pain.
