CYP3A5 RAPA slow metabolizer, asthma stopped in it's tracks

Has anyone had a similar experience with needing a very low dose like 1 mg?

I took RAPA twice, 1 mg for 4 weeks or a total of 4 mg and influenced asthma out of my life.

The next dose 2 months later was a total of 1 mg. (Not 1 mg a week but 1 mg this month).

My lungs are clear. If, rarely i get an attack, my body shuts it down.RAPAis no longer in my system. My lungs or immune system has been rewired.

My immune system was trying to kill me in many ways. But the thing that was destroying my lungs was unrelenting asthma night & morning, 2 hours of coughing to remove mucus each morning and at night, it was out of hand. My lungs were sore, raw.

So I took RAPA. I took 1 mg a week for 4 weeks. After 4 weeks at a total of 4 mg, i was sick from RAPA. I stayed in bed. As the RAPA very slowly left my system, asthma wd flare but within minutes it was quiet. I thought oh, boy, this is great, but this is going to fade, but asthma recovery got stronger. That was April 2026.

Then I took my full genome to Claude AI, fired up Linux on my laptop, ran BWA-MEM bioinformatics, PING (Pushing Immunogenetics to the Next Generation) and went searching for my immune system data. Along the way I found that I lacked a set of genes the CYP3A5 set. My drug clearance hobbled along on one leg, CYP3A4.

I knew nothing about bioinformatics. It was a painful learning curve. It took a month of compute, and mistakes, but I pulled out my immune system, and began doing searches on the genome. I ended up in Google Cloud when my old ThinkPad ran out of memory.

I figured out I had a CYP3A5 *3/*3 gene variant that slowed drug metabolism.

With that information, June 3, I tried RAPA again at 1 mg. I drank 8 ounces of pomegranate juice, i ate a quart of blackberries, and on the heels of that, curcumin supplements, and a half dozen others during the first week that amplified RAPA. Not all supplements do. I got very sick. I went to bed from adding amplifiers. RAPA at 1 mg is okay for me. Amplifiers in my diet duribg the first 8 days are not.

Until then. I thought only grapefruit was an amplifier. It’s the end of June, nearly 30 days have gone by on 1 mg of Rapamycin, and I’m well.

Certainly someone else has had symptoms from RAPA that weren’t due to to the biotanical but to the amplification. I’d like to hear those stories.
