My wife and I are flying down to Orlando tomorrow and then going on a cruise. Should we bump our rapamycin dose up a day early or will we be fine taking it in a checked bag? Has anyone else flown with rapamycin? (We don’t have prescriptions, but have gotten it from India).

Assuming, you only plan to take what you will use yourself on the trip.
Not TSA’s job.
Not a controlled substance.
TSA would have no idea that you don’t have a prescription or a means of determining you don’t
TSA would have no way of determining the Rapamycin wasn’t vitamins.

I’ve been wondering the same thing about taking meds from India with me internationally (Spain). I’m thinking I’ll just take them in their original packaging (blister packets) so it’s at least obvious what they are (empagliflozin and bempedoic acid).

No. They won’t care about your pills. Pack it and bring on the trip. You have absolutely nothing to worry about.
I’ve brought pills all over the world on at least 100 flights.

I’ve packed several thousand pills of prescription drugs (Including over 1,000 tabs of Rapamycin) in my checked luggage from the USA to Hong Kong with no problems.

Just brought some Rapamycin cross country – a small amount, but no problem.
