Adipose tissue (Fat) as a linchpin of organismal ageing

Ageing is a conserved biological process, modulated by intrinsic and extrinsic factors, that leads to changes in life expectancy. In humans, ageing is characterized by greatly increased prevalence of cardiometabolic disease, type 2 diabetes and disorders associated with impaired immune surveillance. Adipose tissue displays species-conserved, temporal changes with ageing, including redistribution from peripheral to central depots, loss of thermogenic capacity and expansion within the bone marrow. Adipose tissue is localized to discrete depots, and also diffusely distributed within multiple organs and tissues in direct proximity to specialized cells. Thus, through their potent endocrine properties, adipocytes are capable of modulating tissue and organ function throughout the body. In addition to adipocytes, multipotent progenitor/stem cells in adipose tissue play a crucial role in maintenance and repair of tissues throughout the lifetime. Adipose tissue may therefore be a central driver for organismal ageing and age-associated diseases. Here we review the features of adipose tissue during ageing, and discuss potential mechanisms by which these changes affect whole-body metabolism, immunity and longevity. We also explore the potential of adipose tissue-targeted therapies to ameliorate age-associated disease burdens.

Iā€™d like to be in the 15-30%. Iā€™m probably there already, so Iā€™d like to get to 15%.

I am at 12%, which is the result of major exercise and diet control. But I have dermal fat around my stomach that is tough to get rid of, so I look more like 15%. Annoying.

My dream is 15% but currently on 25%.
Telmisartan and myo-inositol seem to have a positive effect on body composition through metabolism, visceral fat and things like insulin sensitivity and PPAR gamma.
My question is: would you suggest anything else to add to my ā€˜get sexy bodyā€™ stack.
It currently contains:

  • Telmisartan (although main benefit is for BP and CVD)

  • Myo-inositol

  • MK-677

  • MK-2866 (but I only do a maximum of 12 weeks on followed by tamoxifen PCT

Anything else.

I gym 3 times per week and eat healthy. :joy:

I went from 40-45% to 15-17% and the loose skin phenomenon is real. @desertshores is so right about losing a lot of weight making you look older. My facial age results are 4 years older than my actual age due to this as well and I think surgery might be the only real fix.
That said 6 of my 10 patients Monday were younger than me and 5 of the 6 were morbidly obese so me thinks at the end of the day (putting vanity aside) Iā€™m better off looking a little older yet being fit and healthy.

Hopefully all the hard work and Rapamycin will take you all to where you wish to be. I row (erg) and swim and lift but not as dedicatedly as I should. Between peri menopause and menopause and post menopause it creeps on so as you hardly notice until - oops! 30 percenter! Adiposal! Talk about homonal. @blsm , not sure I look any younger than my 61 years though - whatever ā€œyoungerā€ means!

You are young enough to recover if you can maintain your weight.

I take it for the ghrelin analogue nature. My hypothesis is that part of the benefit from calorie restriction is the grehlin->ghg- igf cascade. I think the ratio of ghg to igf as initiated by ghrelin analogue is life extending i hypothesise ( fingers crossed )

I donā€™t get hungry as I take a fairly low dose at night. The water retention is partly resolved by the ARB (telmisartan)

Thatā€™s very interesting. It made me absolutely ravenous and I gained 7 lbs in two days, but I also have SNPs associated with elevated ghrelin already, so I think it kicked me over the edge.

Retatrutide is a metabolic furnace. Itā€™s worth an experiment.

Retatrutide is on my list.Iā€™ll do my ā€˜due dillingenceā€™ (drug interactions, contraindications, effect on heart, cancer etc.) if all passes my test Iā€™ll experimentā€¦ from my posts youā€™ll see this is very likely.

Iā€™ll add that my eye age came out 10 years younger so itā€™s the rest of my face from losing weight. My latest pheno age based on blood work results was 14 years younger so Iā€™ll just be happy about getting healthier, count my blessings and not stare in the mirror too much. Hahaha

Same here, until I eliminated bread, pasta and cheese from the diet. Then, it literally vanished. No other modifications to diet but over a year period of time. Not easy to refrain from.

Prior to that, I experimented with 7-keto DHEA and found it very effective, but felt turning up the metabolic furnace was not a good idea (subjective decision).
