After 1 week my blood level is 2.4ng/mL. The next day I took 6mg with 2 grapefruit and after 75 minutes it yield 3ng/mL.

I’m waiting for the enteric capsules to arrive and I’ll have these pills inside the capsules and see what happens.

Be careful about your own CYP enzymatic make-up!

About 12 years ago I was on simvastatin and developed community pneumonia. I went to a local clinic. He put me on erythromycin, and never checked what else I was on. Well, simvastatin + erythromycin are strong inhibitors of the CYP3A4. One was fine—two was definitely not.

The next afternoon at work I started feeling horrid and did some research on erythromycin and saw the two really were incompatible. I had a multistrip in my clinic (I’m an RN) and the indicator for blood in urine when solid black, instantly. But there was no blood in my urine. There was however, myoglobin. I had Rhabdomyolysis! Luckily there appears to be no major damage done. Had I kept taking the erythromycin, in a day or three, my liver would have been destroyed.

The pharmacist at CVS said: “The computer should have pinged me when it saw that you were on simvastatin.”

I later found that exactly the same thing happened to my niece. Therefore, I think it’d be wise that I consider going very slowly with the grapefruit juice. Anyone else with similar experience?

The last result is a bit unusual. I had 500ml of GFJ from the bottle. About 1.5hr later I had the 100mg aspirin and some breakfast with my wife consisting of half a garlic bread with avocado. After that on the 2hr mark I had 13x 2mg pills doused in hemp oil. 24.5 hours later I returned a result of 18.2 ng/L. I didn’t even feel tired.

Previously 2 weeks ago, my 24 hour test yield 9.7ng/L. And I felt fatigue on the first day. The only different this time is I had more food, and GFJ from the bottle and took the rapamycin around 10.30am instead of 8am.

Next week I’ll take 12mg encased in enteric capsules blanks I bought and try to repeat what I did this week.

See how I go this week with my blood test. The only question is should I put the hemp oil into the capsules or take them separately? I’m planning to have it separately to maintain the integrity of the capsules.

It seems like a lot of work to replicate the results we would normally get by using Rapammune, Zydus or Biocon brands.

To me, Siroboon isn’t worth the effort.

I think the problem is Australia Border Security not allowing me to order anymore rapamycin until August. Anyway it is interesting and keeps my mind busy lol.

I am glad you keep trying. I have quite a lot of Siriboon and if I get my new supply of Biocon interrupted, it will be my fall-back. My approach was to take a LOT of it, like 20 mg prior to my getting a shipment of Biocon.

Latest result with 12mg in enteric capsule.

In my observation having the pills an hour or 2 after food yields a higher result.

I know someone who takes like 24mg of zydus weekly. Lol

24 mg of Zydus a week is a bit much. He may be inhibiting his MTORC2 at those levels. We do know that levels of 20 mg and below do not typically inhibit MTORC2. At 40 mg, it’s a 50/50 proposition.

I’m tempted to stick with 12mg a week with the enteric capsules. Anyway I’ll check again for the next 2 weeks and decide.

Any side effects? Hopefully the person gets their blood checked… regularly.

apparently that dosage suppresses the psoriasis. If my 12mg does not do anything, I will resort to that.

Just had 12mg 6x2mg earlier today which I put into the enteric capsules. I tried to finish the remainder of the GFJ but it appears to be more than 600ml. It has been 4 hours and I’m actually feeling a bit more tired than usual. These capsule must be working and extra GFJ definitely affects it. Tomorrow I’ll get the 24hr blood test and by Friday I’ll have the results.

Update

24 hour test comes in at 10.1 ng/L

So it seems more GFJ does affect it. That means the peak is over 30+

I think I’ll maintain this level for a few weeks and see how my psoriasis goes and observe of there are any other effects.

The enteric capsules definitely helps. They are very cheap. Like 200 for $15aud. Allows me to reduce from 26mg to 12mg. Eating my scrambled eggs with hemp oil and bread also helps :joy:. From now on I’ll do 1 glass of GFJ 2 to 3 hours before. Then when I have my breakfast an hour before I’ll add another glass of GFJ. I’m sure I’m total more than 500ml of GFJ. GFJ cost $6 per bottle of 1.5L here. I still don’t like them. But for the greater health :joy:. My supply should last 1.5 years at this rate. I’ll extent the weekly to 8 days or longer depending how well the psoriasis behaves. Hopefully I’ll have enough data to convince the local medical institution to allow me to buy locally which is a lot cheaper and higher quality. $30aud per 100 rapamune 1mg.

Either I am getting the dosing wrong or the psoriasis is not diminishing as quickly as I like. Some ppl suggest upping my Vitamin D3 to 3000 IU which I will try. Also, I just got offered 5mg Siroboon tablets for $1.5 each… hmmmm that is cheap. I will only need 2 to be placed into my enteric capsule. Any thoughts?

I wouldn’t bother with Siroboon… their quality just seems far too questionable to bother with. You really have no idea what the dosing is with that company. Many people have reported extremely poor results with their sirolimus. I would only buy Zydus or Biocon because at least you know what dose you’re actually taking.

Just buy Zydus or Biocon from India. Not Siroboon.

I have bought both from India. Although I kind of prefer Zydus.

I have been alternating between zydus and biocon.

I prefer to buy them locally if I can. See what I can come up with. I now have a history of my blood test showing I have been on siromilus so hopefully some unsuspecting GP will prescribe it to me. lol
