Proline restores mitochondrial function and reverses aging hallmarks in senescent cells

i’ve seen you post at longecity. Why don’t you ask Turnbukle for the reference? You can also post that question/request at linkedin (Am not a member).

The article below contains the following statements:

Both mitochondrial fission and fusion seem to be required to maintain mitochondrial function. However, the mechanisms differ. Fission occurs probably to protect function by facilitating equal segregation of mitochondria into daughter cells during cell division and to improve distribution of mitochondria along microtubules tracks. In addition, fission may also help to isolate segments of damaged mitochondria, promoting their clearance by macroautophagy as discussed below [[36](Mitochondrial Fusion/Fission, Transport and Autophagy in Parkinson's Disease: When Mitochondria Get Nasty - PMC)]. Besides to maintain normal mitochondrial functions, mitochondrial fusion, as well as mitochondrial fission have also been associated with cell death/survival mechanisms [37, 38

I am not that focussed on this point. You are welcome to ask yourself.

Okay. I mistakenly thought you were asking me.

Thank you for your reply Juan  Intuitively, I also feel that my crash might point to somewhat dysfunctional mitochondria. I’ll be patient, try some more and see what happens.

Has John or anybody else carried out a high-dose proline trial by now? Belatedly I am thinking about using my big supply of proline.

I took 18 gtams of proline daily for 14 days.

Alas, I was not able to get any mitochondrial biopsy test. A VO2 max test is a very imperfect marker for mitochondrial health since there are other crucial factors in play and the absence of any effect is no proof whatsoever of proline not strengthening mitochondria. I did the test anyway, my VO2 max was up 3 % compared to 18 months ago.

I noticed nothing subjectively, neither positive or negative.

My genetically very high lp(a) which had always been around 200 nmol/L, went down to 124 when I tested it recently. The only radical thing I’ve done during that time is the 14 day 18 g/day proline dose.

The only thing I can find in the literature is small lp()a decreases taking a much longer time to happen with 2-6 g daily doses.

Intriguing. Maybe try a new round of proline mega dosing . Add on lysine and vit C. Minimal evidence, but the upside would be huge.

Juan Daw:

I tested the Louis Dischler protocol, on the tiny chance that there could be something. Results give me a little uncertain hope.

Day 1 100 mg PQQ +4 mg CaAKG (2-3 mg AAKG in the protocol) for mtDNA boost
Day 3 morning 2 g fumaric acid for Up-Latching (Mitochondrial Fusion
fumaric acid to Supersaturate the inner mitochondrial membrane (IMM) by promoting hyperfusion and expansion of the mitochondrial network.

Day 4 morning 1 g citric acid Down-Latching (Mitochondrial Fission phase)

On Day 3 a few hours after the uplatching I managed ten deadlifts at my maximum weight. I was doing five to six before, and a few days later I was down to six again. I do my deadlifts all-out, I can not do a single more why I think there just might be something to the protocol. But of course it could be a very trong placebo effect it was just what i was.hoping for.

Dischler has apparently come up with a new protocol that he claims will make the strenthening from the uplatching more permanent. With the tentative encouragement from his previous protocol I will investigate it. .
