No GFJ does not extend the half-life significantly. The reason Bryans 90 minut level is so much higher than his 4.1 day level is because the 90 minute level is very close to the peak of the biphasic curve. The decline from 90 minutes to 4.1 days does not reflect the half-life accurately because of the biphasic half-life.
When would terminal half life start being measured? I saw a video where Matt Kaeberlein calculated his half life using initial half life (first day so not terminal) so was basing it off of that, but it does make sense to wait until it gets absorbed into the tissues and then test then to get the most accurate blood readings
You’re absolutely right. It makes sense to measure after most of it has been taken up into the tissues. As knowledgeable as Klaberlein is, IIRC he made a critical error in measuring his half-life from measurements taken too soon after dosing. Others pointed this out somewhere on this forum if I remember.
To get an accurate half-life it’s best to wait at least 24 hours to measure blood levels and then measure at a later time point. FYI I measured at 48 and 96 hours and was able to calculate my half-life based on that. It came out very close to what is expected in humans.
