14.7 ppm of Rapamycin = X ppm of Everolimus?

In the ITP they tested the dose 14.7 ppm of Rapamycin. What is the equal dose of Everolimus for that in mice? Let’s together send in a ITP proposal for testing Everolimus. First step is to figure out what dose to test. I have also contacted Joan Mannick and asked her. Deadline for sending in the ITP proposal is 28th of February.

Two reasons why Everolimus could lead to higher lifespan

  1. Shorter half-life (around 28 hours instead of 62 hours as for rapamycin).
  2. Less risk to inhibit mTORC2 and a theory is that avoiding inhibition of mTORC2 is aligned with increased lifespan

These two reasons could also be two good arguments why a higher dose of Everolimus could be tested in other ITP tests to see if it could lead to even higher lifespan effects.

The best lifespan compound we know about today is Rapamycin and I think it’s important that we get one more data on the lifespan effect of the rapalog Everolimus. Why need to get a indication if it has better lifespan potential or not. So let’s put our heads together and create this ITP proposal and try to move the longevity needle.

I asked ChatGPT and got this answer

“The equal dose of everolimus in mice compared to rapamycin is reported to be about 0.3-3 mg/kg for everolimus compared to 1 mg/kg for rapamycin. Here is a PubMed link that supports this: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5649422

The thing that I feel is strange about all this is that it can either be a lower value like 0.3 mg/kg or a very high value like 3mg/kg. It doesn’t make any sense. But I guess it’s the higher value that seems probably the way to go. I hope I get also an answer from Joan Mannick.

I would not trust ChatGPT to be truthful or accurate right now in any area…

This paper seems to cover it:

SRL (sirolimus) mean dose at time of conversion was 2.0 ± 0.9 mg/d. Initial EVL (everolimus) mean dose was 1.3 ± 0.5 mg/d. Six months after conversion
