Dayvigo® (lemborexant), Eisai’s dual orexin receptor antagonist, was approved in China in recently. I am wondering if it’s the safest drug we can get on the market? In the past I tried several other sleep drugs and they don’t work very well on me. I tried a ‘safe’ pill Ramelteon and it halves my deep sleep duration according to the OURA ring. I also tried low dose of trazodone and it made me dr…

I am very interested in sleep quality. At one time in my life I had sleep apnea. When I finally got a good night’s sleep with a CPAP machine I was totally amazed at the difference it made. That was a long time ago but the lesson was learnt and I very much try to have good sleep quality. However I am a bit of a worrier and that can keep me awake. I’ve experimented with a lot of different appro…

I tried lemborexant and find it amazing! In my N=1 experiement, micro-dosing 1mg lemborexant increased my deep sleep according to OURA ring. However I read both lemborexant and rapamycin are motabolized by CYP3A4 enzymes. I’m concerned about potential drug-drug interactions. Is it safe to take rapamycin in the morning and lemborexant at night?

Hi everyone, I’ve been suffered for insomnia for 4 years. Literally I tried all the drugs, therapies and supplements for sleeping improvement. Here I want to record some of my medication history and my own sleeping supplement, since sleeping is one of the three pillars of longevity. Correct me if I make any mistake. Many hormones and neurotransmitters regulate sleep. GABA GABA is the primar…
